We investigated the membrane expression of E- and N-cadherins in three different cell lines used as model systems to study epithelial to mesenchymal transition (EMT) and a possible detection of circulating tumour cells (CTCs). We used various FRET donor-acceptor pairs and protein recognition strategies were utilized, in which Lumi4-Tb terbium complexes (Tb) and different organic dyes were conjugated to several distinct E- and N-cadherin-specific antibodies.

Fluorescent diamond nanocrystals are attracting increasing interest for a broad range of applications. Up to now, diamond nanocrystals have been irradiated as dry powder in a container or deposited as a thin layer on a flat substrate, depending on the type and energy of the irradiating particles. Here, we present an approach based on direct large-scale irradiation of nanodiamonds in aqueous colloidal solution by high-energy protons. This approach results in a larger fraction of fluorescent particles, with a more homogenous distribution of nitrogen-vacancy centers per particle compared to dry powder irradiation.

Distinctive optical properties of inorganic quantum dot (QD) nanoparticles promise highly-valuable probes for fluorescence-based detection methods. Here, a novel bioconjugation strategy is presented that relies on the controlled functionalization of CdSe/CdS/ZnS QD nanoparticles capped with a multidentate dithiol zwitterionic copolymer ligand. We realized a QD-protein A nanoconstructs with controlled stoichiometry and showed that these novel nanobioconjugates are (i) devoid of non-specific interactions, (ii) have a high specificity for extra- and intracellular targets in fixed and live cell experiments, (iii) are colloidally stable and target-biospecific over more than 6 months, and (iv) have very low toxicity after 48 h incubation with living cells.

We report that hydrogenated cationic nanodiamonds of primary size 7 nm (ND-H) have also a high affinity for siRNA and are capable of delivering them in cells with limited toxicity. With siRNA/ND-H complexes, we measured a high inhibition efficacy of EWS/FLI-1 gene expression in Ewing sarcoma cell line.

Multiple frequency electron paramagnetic resonance (EPR) study of small (4–25 nm) nanodiamonds obtained by various dynamic synthesis techniques reveals systematic presence in the half-field (HF) region a distinctive doublet fingerprint consisting of resolved gHF1 = 4.26 and gHF2 = 4.00 signals. This feature is attributed to “forbidden” transitions in EPR spectra of two native paramagnetic centers of triplet (S=1) origin designated as TR1 and TR2.

In this work, we encapsulate bright 30-nm fluorescent nanodiamonds (FNDs) in 10–20-nm thick translucent (i.e., not altering FND fluorescence) silica shells, yielding monodisperse near-spherical particles of mean diameter 66 nm. High yield modification of the shells with PEG chains stabilizes the particles in ionic solutions, making them applicable in biological environments.
